چکیده

Adenosine deaminases acting on RNA (ADARs) have double-stranded binding domains and a deaminase domain (DD). We used the MS2 system specific guide RNAs to direct ADAR1-DD target adenosines in mRNA encoding-enhanced green fluorescence protein. Using this transfected HEK-293 cells, we evaluated effects of changing length position efficiency conversion amber (TAG) ochre (TAA) stop codons tryptophan (TGG) target. Guide 19, 21 23 nt were positioned upstream downstream MS2-RNA, providing total six RNAs. The more functionally effective than RNAs, with following hierarchy efficiency: > 19 nt. highest editing was 16.6%. Off-target not detected complementary region but 50 proportional amount inversely RNA. Our results suggest that requires precise optimization ratio enzyme, RNA,

Editing of HIV-1 RNA by the double-stranded RNA deaminase ADAR1 stimulates viral infection

Adenosine deaminases that act on dsRNA (ADARs) are enzymes that target double-stranded regions of RNA converting adenosines into inosines (A-to-I editing) thus contributing to genome complexity and fine regulation of gene expression. It has been described that a member of the ADAR family, ADAR1, can target viruses and affect their replication process. Here we report evidence showing that ADAR1 ...

Modulation of ADAR1 editing activity by Z-RNA in vitro

RNA editing by A-to-I modification has been recognized as an important molecular mechanism for generating RNA and protein diversity. In mammals, it is mediated by a family of adenosine deaminases that act on RNAs (ADARs). The large version of the editing enzyme ADAR1 (ADAR1-L), expressed from an interferon-responsible promoter, has a Z-DNA/Z-RNA binding domain at its N-terminus. We have tested ...

Serotonin-2C receptor pre-mRNA editing in rat brain and in vitro by splice site variants of the interferon-inducible double-stranded RNA-specific adenosine deaminase ADAR1.

The interferon-inducible RNA-specific adenosine deaminase (ADAR1) is an RNA editing enzyme implicated in the site-selective deamination of adenosine to inosine in cellular pre-mRNAs. The pre-mRNA for the rat serotonin-2C receptor (5-HT2CR) possesses four editing sites (A, B, C, and D), which undergo A-to-I nucleotide conversions that alter the signaling function of the encoded G-protein-coupled...

RNA Editing, ADAR1, and the Innate Immune Response

RNA editing, particularly A-to-I RNA editing, has been shown to play an essential role in mammalian embryonic development and tissue homeostasis, and is implicated in the pathogenesis of many diseases including skin pigmentation disorder, autoimmune and inflammatory tissue injury, neuron degeneration, and various malignancies. A-to-I RNA editing is carried out by a small group of enzymes, the a...
